Expression of Flavobacterium proteins for bioscience and vaccines

During a study aimed at identifying enzymes with optimal activity at low temperatures, we identified several members of the phylum Bacteroidetes as interesting sources for new enzymes. At the early stage of discovery, a heterologous expression system that can provide high yields of enzyme for evaluation is very helpful.
In parallel to the discovery of new enzymes, we developed a lactococcal expression system (designated P170) that reduces the time used on down-stream processing.
Surprisingly, this Gram-positive system proved to be highly efficient to produce enzymes with optimal activity at low temperatures. Moreover, protein candidates with clinical relevance were secreted by the lactococcal host that is regarded as safe and does not accumulate endotoxins.
In this communication we will present the results of the P170 expression system in (i) the expression of a low temperature functioning enzyme and (ii) the production of a vaccine candidate for human administration.
