Optimization Run
In order to obtain the best possible results the optimal level of the following parameters will be established: proteinase treatment, hybridization temperature, and the probe concentration. The run will include control probes: a positive control LNA™ probe detecting snRNA U6 that provides a rough evaluation of the RNA content in the samples. A positive control microRNA LNA™ probe specific for a known microRNA that provides documentation whether the samples allows detection of microRNA. A negative control LNA™ probe, which is a standard scrambled probe with no homology to known transcripts. Interpretation of the results from the Optimization Run allows a firm conclusion based on consideration of the level of uniformity and reproducibility of the specific signal and identification of the best samples for follow up studies: Block Screening, Probe Screening, or Fluorescence Applications.
